Gentamicin and other cassettes for chromosomal gene replacement in Escherichia coli.
Small genetic elements conferring resistance to antibiotics are widely used for strain construction in bacteria. Here, we describe a gentamicin resistance cassette with properties optimized for efficient gene replacement in Escherichia coli. This cassette offers advantages in versatility and ease of use over previously described versions. The Red recombination system of bacteriophage λ, in the ...
متن کاملEngineering of promoter replacement cassettes for fine-tuning of gene expression in Saccharomyces cerevisiae.
The strong overexpression or complete deletion of a gene gives only limited information about its control over a certain phenotype or pathway. Gene function studies based on these methods are therefore incomplete. To effect facile manipulation of gene expression across a full continuum of possible expression levels, we recently created a library of mutant promoters. Here, we provide the detaile...
متن کاملFunctional gene cassettes in development.
It has been noted in recent years that groups ofgenes are often used at multiple stages and multiple places of development. Comparison of the functions of each group of genes in different developmental context gave rise to the notion of "functional gene cassettes." These groups of genes can often be viewed as sets of tools; each group is suited to carry out a certain operation and may serve sim...
متن کاملIntegrons and gene cassettes in the enterobacteriaceae.
Integrons were detected in 59 of 120 (49%) urinary isolates of Enterobacteriaceae by PCR using degenerate primers targeted to conserved regions of class 1, 2, and 3 integrase genes. PCR sequencing analysis of the cassette arrays revealed a predominance of cassettes that confer resistance to the aminoglycosides and trimethoprim.
متن کاملPromoters and control elements: designing expression cassettes for gene therapy.
It has become apparent that the clinical success anticipated in the field of gene therapy has been limited by progress in several of the fundamental areas of genetics, molecular and cellular biology relevant to its application. Whilst a great deal of effort has been made in the evaluation of transgenes, it is only more recently with the advance of vector systems that attention has begun to be f...
